Anatomical Studies by Paraffin Section Method and Subculture on the Anther Callus of Vegetable Soybean

Anatomical studies by paraffin section method for anther callus of vegetable soybean showed that there were four anther sac which connected with connective tissue, The cells in connective tissue, anther wall and pollen are able to divide to form callus. But we could hardly recognize the callus come from which kind of cells. Therefore, it is necessary to examine the chromosome number in root-tip cell when the callus regenerated to plantlet. The suitable medium for subculture of anther callus of vegtable soybean was MS basic medium with 5 mg/L Kinetin, 0.05 mg/L IAA and coconut water 150 ml/L.
